combination effect of gstm1, gstt1 and gstp1 polymorphisms and risk of systemic lupus erythematosus
نویسندگان
چکیده
background : progression of systemic lupus erythematosus (sle) could be due to oxidative stress especially through reactive oxygen species (ros). detoxification of ros is largely performed by glutathione s-transferases (gsts), therefore polymorphisms of gstm1, gstt1 and gstp1 genes which decrease enzymes activity could affect sle susceptibility. the aim of this study was to determine the effects of gstm1 (deletion), gstt1 (deletion) and gstp1 (ile105val) polymorphisms on sle susceptibility. methods : genomic dna was extracted from blood samples of 163 sle patients and 180 age, sex and ethnically matched controls. gsts genotypes were determined by polymerase chain reaction (pcr)-multiplex procedure or pol-ymerase chain reaction-restriction fragment length polymorphism (pcr-rflp) analysis. results : gstt1 null genotype frequency was higher in sle patients than controls. no association observed between gstm1 null genotype or gstp1 ile105val polymorphism with sle. nevertheless combination of gstt1 null/ gstm1 null genotypes showed 2.8-fold increase in risk of sle. moreover the combination of gstt1 null/ gstm1 null/gstp1 ile/val and val/val genotypes increased the sle risk about 8 fold. conclusion : present data suggest that gstt1 null/ gstm1 null/gstp1 ile/val and val/val genotypes might large-ly contribute to the pathogenesis of sle.

Combination Effect of GSTM1, GSTT1 and GSTP1 Polymorphisms and Risk of Systemic Lupus Erythematosus
BACKGROUND Progression of systemic lupus erythematosus (SLE) could be due to oxidative stress especially through reactive oxygen species (ROS). Detoxification of ROS is largely performed by Glutathione S-transferases (GSTs), therefore polymorphisms of GSTM1, GSTT1 and GSTP1 genes which decrease enzymes activity could affect SLE susceptibility. The isoenzymes of the glutathione s transferase (GST) family play a vital role in phase II of biotransformation of many substances. Using a multiplex polymerase chain reaction and a direct sequencing analysis, the frequencies of GSTM1, GSTT1, and GSTP1 polymorphisms were evaluated in 1,051 Korean male subjects. We found that 53.8% of the individuals had the GSTM1 null genotype and 54.3% had the...
متن کاملArsenic Methylation, GSTT1, GSTM1, GSTP1 Polymorphisms, and Skin Lesions
OBJECTIVE We investigated whether primary and secondary arsenic methylation ratios were associated with skin lesions and whether GSTT1, GSTP1, and GSTM1 polymorphisms modify these relationships. METHODS A case-control study of 600 cases and 600 controls that were frequency matched on age and sex was conducted in Pabna, Bangladesh, in 2001-2002. Individual well water, urine, and blood samples ...
متن کاملGenetic Polymorphisms of Glutathione-Related Enzymes (GSTM1, GSTT1, and GSTP1) and Schizophrenia Risk: A Meta-Analysis
The association between polymorphisms of glutathione-related enzyme (GST) genes and the risk of schizophrenia has been investigated in many published studies. However, their results were inconclusive. Therefore, we performed a meta-analysis to explore the association between the GSTM1, GSTT1, and GSTP1 polymorphisms and the risk of schizophrenia. Twelve case-control studies were included in thi...
متن کاملGSTM1, GSTT1, and GSTP1 polymorphisms and risk of advanced colorectal adenoma.
Cigarette smoking is a risk factor for colon adenoma. The glutathione S-transferase enzymes are involved in the detoxification of carcinogenic compounds including those found in tobacco smoke, and thus, may be important modifiers of individual risk of developing this disease. We examined the prevalence of GSTM1 and GSTT1 gene deletions, and two GSTP1 polymorphisms in 772 cases with advanced col...
